Аннотация: Background: The high prevalence of chronic heart failure (CHF) in industrialized countries,&#xD;
alongside its grave consequences—including increased mortality, disability, functional&#xD;
impairment, and reduced quality of life—underscores the urgent need to optimize&#xD;
core medical therapy. In this regard, metabolic cardioprotectants are of particular interest,&#xD;
as their action is aimed at interrupting or mitigating the cascade of adverse metabolic reactions&#xD;
and enhancing the compensatory and adaptive mechanisms of the heart. The aim&#xD;
of this study was to evaluate the effect of sulfur-containing pharmacological agents—&#xD;
namely, N-acetylcysteine (NAC), Heptral, Thiotriazoline, and Angiolin—on selected parameters&#xD;
of the NO and thiol-disulfide systems in the hearts of rats with experimental&#xD;
doxorubicin-induced CHF, followed by an assessment of their cardioprotective efficacy</summary>

Аннотация: Background: The management of pharmacotherapy in patients with&#xD;
multimorbidity and polypharmacy is a difficult task in routine clinical practice.&#xD;
Clinicians must consider diagnoses, prescribed drugs, contraindications, drugdrug&#xD;
interactions, renal and hepatic function, laboratory values, dose limits&#xD;
and individual risk factors. Clinical decision support systems in traditional&#xD;
settings are based on static rules, and hard to update. Standalone large&#xD;
language models are capable of processing clinical language but their outputs&#xD;
may be unsupported by evidence, incomplete or out of date. In this study, we&#xD;
developed and evaluated a hybrid multi-agent GraphRAG framework for&#xD;
personalised pharmacotherapy safety verification.&#xD;
Methods: We developed a system that integrates graph based retrieval with large&#xD;
language models. The framework was composed of three parts: (1) a schema for&#xD;
a pharmacotherapy knowledge graph that encoded indications,&#xD;
contraindications, interactions, laboratory thresholds, dose limitations and&#xD;
relevant patient conditions; (2) an Extractor–Critic workflow that transformed&#xD;
unstructured medical text into graph elements with quality checks prior to&#xD;
their incorporation; and (3) a retrieval and re-ranking module that leveraged&#xD;
semantic search, graph traversal and safety-directed scoring. The system was&#xD;
evaluated on 12 benchmark cases extraction and 30 clinical safety questions&#xD;
answering. Comparison of conventional RAG and GraphRAG was done using&#xD;
linear mixed effects models, permutation testing, and paired case-level analysis.</summary>

Аннотация: Actuality. Intranasal drug delivery is considered a promising approach to enhancing the efficiency of neuroprotective therapy.&#xD;
However, long-term use of such drugs requires a comprehensive assessment of its safety.&#xD;
The aim of the study was to evaluate the chronic toxicity of 1% Angiolin nasal gel following its prolonged administration.&#xD;
Materials and methods. The study was conducted on white non-linear rats following intranasal administration of 1% Angiolin gel&#xD;
at a dose of 100 mg/kg for 180 days. The general condition of the animals, behavioral responses (Open Field test), indicators of cardiac&#xD;
bioelectrical activity, hematological and biochemical parameters of blood and urine, and the morphological condition of organs were&#xD;
assessed. Immunotoxic effects were studied based on the weight and cell count of lymphoid organs, peripheral blood, and humoral and&#xD;
cellular immune responses under conditions of normal immune status and cyclophosphamide-induced immunodeficiency. Актуальність. Інтраназальна доставка лікарських засобів розглядається як перспективний підхід підвищення ефектив-&#xD;
ності нейропротективної терапії. Однак тривале застосування таких препаратів потребує комплексної оцінки їхньої безпеки.&#xD;
Мета дослідження. Оцінка хронічної токсичності 1%-го назального гелю Ангіолін за тривалого введення.&#xD;
Матеріали та методи. Дослідження виконано на білих нелінійних щурах за інтраназального введення 1%-го гелю Ангіо-&#xD;
лін у дозі 100 мг/кг протягом 180 діб. Оцінювали загальний стан тварин, поведінкові реакції (тест «Відкрите поле»), показ-&#xD;
ники біоелектричної активності серця, гематологічні та біохімічні параметри крові й сечі, морфологічний стан органів.&#xD;
Імунотоксичну дію вивчали за масою та клітинністю лімфоїдних органів, показниками крові, гуморальною та клітинною&#xD;
імунною відповіддю, в умовах нормального імунного статусу та циклофосфан-індукованому імуноімунодефіциту.</summary>
