Аннотация: В дослідженні вивчались NO-залежні механізми кардіопротективної дії МТ в умовах інфаркту міокарда. Встановлено, що призначення МТ (розробка НВО «Фарматрон») у дозі 2,5 мг/кг внутрішньоочеревинно паралельно формуванню експериментального гострого інфаркту міокарда приводило до підвищення продукції NO в цитозолі і мітохондріях ішемізованого міокарда за рахунок підвищення експресії і активності ендотеліальної NO-синтази. Введення МТ сприяє також підвищенню біодоступності цього молекулярного месенджера, підсилюючи протективну дію відновлених тіолів. Призначення МТ значно обмежує прояви нітрозуючого стресу в ішемізованому міокарді. Застосування МТ за силою терапевтичної дії достовірно перебільшує ефективність референс-препарату - метопрололу.</description>

Аннотация: Background: The high prevalence of chronic heart failure (CHF) in industrialized countries,&#xD;
alongside its grave consequences—including increased mortality, disability, functional&#xD;
impairment, and reduced quality of life—underscores the urgent need to optimize&#xD;
core medical therapy. In this regard, metabolic cardioprotectants are of particular interest,&#xD;
as their action is aimed at interrupting or mitigating the cascade of adverse metabolic reactions&#xD;
and enhancing the compensatory and adaptive mechanisms of the heart. The aim&#xD;
of this study was to evaluate the effect of sulfur-containing pharmacological agents—&#xD;

Аннотация: Background: The management of pharmacotherapy in patients with&#xD;
multimorbidity and polypharmacy is a difficult task in routine clinical practice.&#xD;
Clinicians must consider diagnoses, prescribed drugs, contraindications, drugdrug&#xD;
interactions, renal and hepatic function, laboratory values, dose limits&#xD;
and individual risk factors. Clinical decision support systems in traditional&#xD;
settings are based on static rules, and hard to update. Standalone large&#xD;
language models are capable of processing clinical language but their outputs&#xD;
may be unsupported by evidence, incomplete or out of date. In this study, we&#xD;
developed and evaluated a hybrid multi-agent GraphRAG framework for&#xD;
personalised pharmacotherapy safety verification.&#xD;
Methods: We developed a system that integrates graph based retrieval with large&#xD;
language models. The framework was composed of three parts: (1) a schema for&#xD;
a pharmacotherapy knowledge graph that encoded indications,&#xD;
contraindications, interactions, laboratory thresholds, dose limitations and&#xD;
relevant patient conditions; (2) an Extractor–Critic workflow that transformed&#xD;
unstructured medical text into graph elements with quality checks prior to&#xD;
their incorporation; and (3) a retrieval and re-ranking module that leveraged&#xD;
semantic search, graph traversal and safety-directed scoring. The system was&#xD;
evaluated on 12 benchmark cases extraction and 30 clinical safety questions&#xD;
answering. Comparison of conventional RAG and GraphRAG was done using&#xD;
linear mixed effects models, permutation testing, and paired case-level analysis.</description>

Аннотация: Новий антиангінальний препарат «Гіпертрил» (МТ) (похідне 4-аміно-1,2,4-тріазолу), створений на НВО «Фарматрон», за рахунок кардіопротективних механізмів, скерованих на зниження як перед-, так і постнавантаження на серце, покращує показники системної і кардіогемодинаміки в умовах гострої ішемії. Введення гіпертрилу кролям з гострою ішемією міокарда дозою 2,5 мг/ кг внутрішньовенно зумовлює сповільнення серцевого ритму, зниження артеріального тиску і зменшення загального периферичного опору судин. Гіпертрил відновлює функції лівого шлуночка, що виражається в збільшенні робочого індексу лівого шлуночка і робочого ударного індексу лівого шлуночка, підвищенні тиску в лівому шлуночку на 20%. Гіпертрил спричинює збільшення ударного об’єму, систолічного та серцевого індексів. Кардіопротективна дія референс-препарату «Метопролол» (10 мг/кг) зумовлена переважно зменшенням переднавантаження на серце. New antianginal medication «Gipertril» (MT) (derivative of 4-amino-1,2,4-triazole), product of Scientific Production Association “Pharmatron” (president - professor I.A. Mazur) due to cardioprotective mechanisms directed on the decrease of both preload and post-load on heart improves indexes of the system hemodynamics and heart hemodynamics in the conditions of acute myocardial ischemia (AMI). AMI was induced in outbred rabbits by intravenous injection of caffeine - sodium benzoate solution in dose 25 mg/kg and then 3 minutes later adrenaline hydrochloride was introduced intravenously at the rate of 0,2 mg of pure adrenaline per kg. Intravenous introduction of «Gipertril» to the rabbits with AMI in the dose of 2,5 mg/kg 30 min before introduction of caffeine - benzoate resulted in significant decrease of general peripheral vascular resistance (GPVR) by 30% as compared with the control group that testifies to vasodilating effect of this medication. Introduction of metoprolol as opposed to «Gipertril» increased GPVR in animals with AMI. In groups of animals, which received «Gipertril» and metoprolol, decrease of heart rate was noted that is the medication showed negative chronotropic effect typical of all P-adrenoceptor antagonists, decreasing pre-load on heart in the period of acute ischemia. In the group which received «Gipertril» heart rate was significantly lower by 27% over all period of the experiment as compared with the control group.</description>
