Аннотация: Метою нашої роботи є синтез нових сполук – похідних 4-аміно-5-(4- третбутилфеніл)-4Н-1,2,4-триазол-3-тіолу, дослідження їхніх протимікробних властивостей та встановлення деяких закономірностей залежності «структура – дія». Wide and uncontrolled use of antibiotics and antimicrobial drugs among the population led to increase of resistance of microorganisms. So even slight infections and injuries can lead to serious consequences. Analysis of the current literature of organic synthesis showed that one of the most promising classes for synthesis of new drugs is the class of 1,2,4-triazole. This class of chemical compounds is well established in clinical practice owing to wide spectrum of biological activities (antimicrobial, hypotensive, antispasmodic, etc.) and a small amount of side effects. The aim of the work is the synthesis of new compounds – derivatives of 4-amino-5-(4-tertbutylphenyl)- 4H-1,2,4-triazole-3-thiol, investigation of their antimicrobial properties and establishment certain patterns of dependence «structure–action». Objects of investigations were the salt – the derivatives of 4-amino-5-(4-tertbutylphenyl)-4H-1,2,4- triazole-3-thiol. During the work elemental analysis, chromatography-mass-spectrometric investigation, 1H-NMR spectroscopy, the methods of synthesis and were carried out and was investigated the antimicrobial activity of the test compounds. Researches have established some dependence «structure–action» and have identified compounds whose antimicrobial activity exceeded the indicators of the reference preparation – chlorhexidine. In the result of researches original methods of synthesis derivatives of 4-amino-5-(4-tertbutylphenyl)-4H- 1,2,4-triazole-3-thiol was suggested, promising compounds which exhibit an antimicrobial action was revealed and some regularity «structure–action» was installered. Матеріали та методи дослідження. Експериментальну гіперліпідемію моделювали на 20 білих нелінійних щурах введенням холестерину та ергокальциферолу. Об’єктом дослідження були мітохондріальна та цитозольна фракції гомогенату серця. Визначали активність НАД-залежної малатдегідрогенази (НАД-МДГ), сукцинатдегідрогенази (СДГ), цитозольної глутатіонпероксидази (ГПО), рівень нітротирозину (методом ІФА) та ступінь відкриття гігантських мітохондріальних пор (МП). Препаратом порівняння слугував аторвастатин. The aim of the study – to evaluate the effect of the original lead compound, sodium 2-((4-phenyl-5-(thiophen-3-ylmethyl)-4H- 1,2,4-triazol-3-yl)thio)acetate, on mitochondrial functional state and markers of nitrosative-oxidative stress in the rat heart under experimental hyperlipidemia. Research methods. Experimental hyperlipidemia was induced in 20 white outbred rats by the administration of cholesterol and ergocalciferol. The mitochondrial and cytosolic fractions of heart homogenate were the objects of the study. The activities of NADdependent malate dehydrogenase (NAD-MDH), succinate dehydrogenase (SDH), and cytosolic glutathione peroxidase (GPx) were determined. The levels of nitrotyrosine (via ELISA) and the mitochondrial permeability transition pore (mPTP) opening degree were also measured. Atorvastatin served as the reference drug. В умовах збройної агресії рф проти України система охорони здоров’я зазнала суттєвих трансформацій, що спричинило виникнення нових бар’єрів у доступності лікування та підтримці хворих на РС. Фармацевтичний складник у цих умовах набуває критичного значення для забезпечення раціональної терапії та підтримки прихильності пацієнтів до лікування. Мета – дослідити ключові проблеми медико-фармацевтичної допомоги пацієнтам із РС в Україні в умовах воєнного стану та визначити перспективні напрями посилення ролі фармацевтів у системі їхнього медичного супроводу. Матеріали та методи дослідження. Проведено анкетне опитування 121 пацієнта з підтвердженим діагнозом РС із різних регіонів України. Для аналізу використано методи описової статистики, узагальнення та систематизації даних за допомогою програмного забезпечення Microsoft Excel. Опитування охоплювало блоки питань щодо доступності діагностики, лікування, реабілітації та фармацевтичного супроводу. Multiple sclerosis (MS) is a progressive autoimmune disease that requires continuous medical and pharmaceutical care. In the context of the armed aggression of the Russian Federation against Ukraine, the healthcare system has undergone significant transformations, which has led to the emergence of new barriers in the availability of treatment and support for MS patients. The pharmaceutical component in these conditions is of critical importance for ensuring rational therapy and maintaining patient adherence to treatment. The aim is to investigate the key problems of obtaining medical and pharmaceutical care by MS patients in Ukraine under martial law and to identify promising areas for strengthening the role of pharmacists in the system of their medical support. Materials and methods of the study. A questionnaire survey of 121 patients with a confirmed diagnosis of MS from different regions of Ukraine was conducted. For the analysis, descriptive statistics, generalization and systematization of data using Microsoft Excel software were used. The survey covered blocks of questions on the availability of diagnostics, treatment, rehabilitation and pharmaceutical support.
